Development of swine immune reagents for swine immunity and biomedical research

Abstract The USDA-NIFA Swine Immune Toolkit Initiative has been involved in generating priority immune reagents for swine immunity and biomedical research pipelining them marketing. With the help of commercial partners we expressed each target molecule as a soluble protein using yeast expression systems then produced panels monoclonal antibodies (mAbs) against target. Most recently generated mAbs reactive with porcine IL-5, IL-6, IL-13, IL-21, IL-28B, CXCL10, BAFF screened their reactivity multiple assays. We determined mAb orthologous proteins most mAbs. A sensitive sandwich ELISA is now available IL-13 CXCL10; other targets are being best pairs. Reactivity tests labeled α-CXCL10, α-IL-6 α-IL-13 intracellular staining cells flow cytometry assays underway different cell stimulation conditions. Immunohistochemistry analyses binding α-CXCL10 on formalin fixed pig lymph nodes spleen tissues progress. Further, analysis large panel α-human CD antigen cross-reactivity ongoing. For target, our goal to provide veterinary community new standardized assay techniques efforts. Finally, efforts proceeding develop SLA tetramers presentation studies.
